Twenty-four well tissue culture plate containing proliferating early passage normal human endothelial cells co-cultured with early passage normal human interstitial cells in a specially formulated culture medium. These cultures spontaneously develop a network of capillary-like tubules after 10-14 days in culture. Tubule development is believed to closely mimic in vivo angiogenesis and is enhanced by known stimulators of angiogenesis and suppressed by known angiogenesis inhibitors, both in a dose-dependent manner.

The kit contains sufficient culture medium to support development of the assay over the required 14 day period.

In summary:

  • Co-culture of primary human endothelial cells with primary human interstitial cells
  • No other additions or manipulations except addition of test substances and medium changes
  • Kits are shipped at ambient temperature ready to use
  • Anastomosing tubule network forms within 10-14 days
  • Tubules stained with antibodies to von Willebrand factor (Fig 1c), PECAM-1 or collagen type IV (1a)
  • Permits evaluation of both inhibitors and stimulators of angiogenesis
  • 24 well format - compatible with replicate samples or secondary screening needs
  • Several approaches to analysis of results are available that include tubule length, branch points, tubule area in parallel to angiogenic inhibitor & stimulator specific immunoassays
